Modified Method for the Estimation of Plasma Ammonia-N by Ion-exchange


Abstract
It has been found possible to simplify the technique and make improvements to both reagents and apparatus with the experience gained in routine daily use of the method by ionexchange. The purpose of this account is to describe the modified method and to mention precautions which are necessary to obtain reliable results. With certain modifications in apparatus and technique, care in the preparation and preservation of reagents have eliminated the difficulties from plasma ammonia analysis.
A study has been made of indophenol blue reaction and modifications of Fenton¢¥s method. are proposed which increase sensitivity as well as accuracy.
The addition of sodium hypochlorite solution of PH range of 12-13 to an ammonia solution before the addition of sodium phenate has been shown to increase the sensitivity and accuracy of the indopheriol blue reaction.
The available hypochlorite concentrations are 0. 01-0. 02 N. respectively. The use of this concentration of hypochlorite solution will result in the maximum color . being produced in addition of alkaline phenate solution after 3-minutes and this is approximately 18% greater than that produced by Fenton¢¥s method.
The effect of temperature and pH of the phenate solution on the reaction are the same by our modified method when the 0.01-0.02N hypochlorite is used. By increasing the sensitivity of the method and eliminating the need for the very rapid addition of reagent, the use of this concentration of hypochlorite results in the most sensitive method.
This modified method may be applied to the later stages and apparatus of Fenton¢¥s method of estimating plasma ammonia and while it increases the sensitivity and accuracy, together with simplify the technique it does not impair the specificity.
